Purpose Potentially toxic element (PTE) concentrations in mine-degraded soil and their bioaccumulation in food crops is a public health concern worldwide. The current study investigates the influence of organic geo-sorbents including biochar (B), farmyard manure (FYM), and peat moss (PTM) on PTE in chromite mine-degraded soil and their subsequent effects on spinach biomass, PTE uptake, average dietary intake (ADI), and health risk (HRI) associated with PTE via spinach consumption.Materials and methods Chromite mine-degraded soil samples were collected from different mining sites in Kohistan region. Pot experiments were carried out in the greenhouse environment. The selected geo-sorbents (B, FYM and PTM) were mixed at application rates of 1%, 2%, and 5%. Contaminated soil without geo-sorbents (control treatment) was also included in each batch of the experiments. Local FYM and PTM were used in this experiment, while B was provided by the Institute of Urban Environment (CAS) Xiamen, China. The total carbon (C), total nitrogen (N), and total sulfur (S) contents in mine-degraded soil and organic geo-sorbents were measured using a macro-elementor (VarioMax CNS, Germany). Total (acid digestion) and bioavailable PTE (As, Cd, Cr, Ni, Zn, and Pb) concentrations in mine-degraded soil and spinach were determined using inductive coupled plasma mass spectrophotometer (ICP-MS 7500 CX, Agilent Technologies, USA).Results and discussion The addition of organic geo-sorbents effectively immobilized the PTE concentrations in mine-degraded soil, and increased the major nutrient contents and thereby reduced the bioaccumulation of PTE (Cr, As, Ni, Cd, Zn, and Pb) in spinach. Consequently, B2, B5, FYM2, FYM5, PTM2, and PTM5 amendments significantly (p<0.001) increased the biomass, whereas the B1, FYM1, and PTM1 addition showed no significant increase in spinach biomass as compared to the control treatment. The results showed that all the organic geo-sorbents had significantly (p<0.001) reduced the As uptake in spinach, while B2, B5, FYM2, FYM5, and PTM5 significantly (p<0.001) decreased PTE bioaccumulation as compared to the control treatment.Conclusions The highest application rate (5%) showed the best result in increasing the spinach growth and biomass as well as reducing the PTE mobility in soil, and their bioaccumulation in spinach, as compared to 1% and 2% application rates and also with the control treatment. Furthermore, the average dietary intake (ADI) of PTE and health risk indices (HRIs) reduced via spinach consumption for both the children and adults, due to the addition of selected organic geo-sorbents used for soil amendments.

Cognitive decline in dementia is associated with deficiency of the cholinergic system. In this study, five mono-carbonyl curcumin analogs were synthesized, and on the basis of their promising in vitro anticholinesterase activities, they were further investigated for in vivo neuroprotective and memory enhancing effects in scopolamine-induced amnesia using elevated plus maze (EPM) and novel object recognition (NOR) behavioral mice models. The effects of the synthesized compounds on the cholinergic system involvement in the brain hippocampus and their binding mode in the active site of cholinesterases were also determined. Compound h2 (p < 0.001) and h3 (p < 0.001) significantly inhibited the cholinesterases and reversed the effects of scopolamine by significantly reducing TLT (p < 0.001) in EPM, while (p < 0.001) increased the time exploring the novel object. The % discrimination index (DI) was significantly increased (p < 0.001) in the novel object recognition test. The mechanism of cholinesterase inhibition was further validated through molecular docking study using MOE software. The results obtained from the in vitro, in vivo and ex vivo studies showed that the synthesized curcumin analogs exhibited significantly higher memory-enhancing potential, and h3 could be an effective neuroprotective agent. However, more study is suggested to explore its exact mechanism of action.

Purpose Antibiotics are growing environmental contaminants leading to public health concern. Antibiotics are commonly used as growth promoters and therapeutic agents in poultry feed that are not completely metabolized in the body tissues of chicken, get deposited in meat as parent compounds, and ultimately excreted via poultry droppings into the environment. These antibiotics in the soil result into the creation of antibiotic resistance in bacteria via activation of antibiotic resistance genes (ARGs). The development of ARGs and antibiotic-resistant bacteria (ARB) lead to huge physical and economic losses, as these bacteria cannot be treated with commonly used antibiotics. Moreover, these antibiotics after entering into food chains seriously affect the human immune system, growth, and metabolism of the body. Therefore, to reduce the future health risks of antibiotics, there is a dire need to understand the fate of poultry antibiotics and spread of ARGs in the soil environment.Materials and methods In this manuscript, we reviewed the existing literature about the antibiotics used in the poultry sector, soil contamination through application of poultry manures, and development of ARB in environment. An attempt has been done to present a better understanding of emerging contaminants (ARGs, ARB) in the soil environment and their associated human health effects.Results and discussion In this paper, we summarized the use of antibiotics in the poultry sector, persistence of antibiotics in animal body, and their release into environment. Transfer mechanism of antibiotics and their metabolites to the human body and their fatal effects have been investigated. Developments of ARB and ARGs in the soil due to excessive use of veterinary antibiotics have been highlighted.Conclusions Poultry antibiotics are causing human health risks by development of ARGs and ARB. Such antibiotic resistance cannot be treated with common antibiotics. Therefore, effective measures are needed to control this emerging problem by improving the efficiency of antibiotics, reducing the spread of resistance genes, and proper monitoring of antibiotics in poultry feed and manure. Manure composting and biochar application are the possible ways to reduce the risk and spread of ARGs in environment due to manure application in agriculture field. The pathways that allow antibiotic, ARGs, and ARB to move through the environment are not fully understood and there is a need for further research to make clear the reservoirs and routes of antibiotic-related contaminants in the ecosystem.

Vigorous knowledge on the occurrence and distribution of toxic metals (TMs) in theenvironment is needed to assess their toxicological impacts on human health especially in developing countries like Pakistan. For the first time, the concentrations of TMs like Ni, Zn, Pb, Cr, and Cd in both ground and surface water and their potential health risk in the district Buner (Pakistan) were determined using Inductive Coupled Plasma Mass Spectrometry (ICP-MS, Agilent Technologies, 7500 CX, USA) and their results were compared with their safe limits defined by Pakistan Environmental Protection Agency (Pak-EPA) and World Health Organization WHO (2008). The concentrations of TMs, like Pb (43% and 86%), Ni (63% and 32%), Cr (41%), and Cd (1% and 20%) in drinking water samples exceeded their respective permissible limits set by Pak-EPA and WHO (2008), respectively. The mean concentrations of TMs were found in decreasing order Ni > Pb > Cr > Cd > Zn. The HRI values of TMs in drinking water samples for both adults and children were observed <1 indicating no health risk to the local population. Excessive marble industries are present in the study area; therefore, it is necessary to reduce metal contamination via proper disposal and treatment of marble wastewater, for which the government should take serious action in the study area.

The study aimed to assess the physicochemical parameters (pH, electrical conductivity (EC), total dissolve solid (TDS), oxidation reduction potential (ORP), Temperature) and potential toxic metals (PTMs), including Ni, Mn, Cr, Cu, Cd, Pb, Co, Fe and Zn in the groundwater of Lower Dir, Pakistan. Furthermore, the pollution sources and spatial distribution pattern of PTMs were also investigated via principal component analysis (PCA) and geographic information system (GIS) application to understand the changing behaviors of PTMs in groundwater. The average concentrations of physicochemical parameters such as pH, EC, TDS, ORP and Temperature were 7.1, 418 mu S/cm, 251 mg/L, 193 mV and 25.7 degrees C, while the concentrations of PTMs; Ni, Mn, Cr, Cu, Cd, Pb, Co, Fe and Zn were 0.25, 0.34, 0.09, 0.29, 0.10, 0.08, 0.10, 0.83 and 0.25 mg/L, respectively. Among the selected metals, Mn, Cr, Cd, Pb, Co and Fe were exceeded the WHO guidelines and their percentage contribution were 43%, 57%, 45%, 70%, 70% and 62%, respectively. The increasing order of PTMs were; Pb > Co > Fe > Cr > Cd > Mn > Cu > Ni > Zn in the study area. PCA represented three significant factors, which explained 76% variability in the groundwater. Whereas, clustering analysis (CA) grouped groundwater into three distinct clusters less polluted (C1), moderate polluted (C2) and highly polluted (C3). Human health risk assessment was carried out to check the suitability of groundwater for drinking and domestic uses. The HQ and HRIs values of Cd were >1, suggested that the groundwater sources are unfit for drinking and domestic purposes and may be caused potential health risk after long term ingestion.
